Gene Summary

Gene Symbol
Masp1

Model Description

The MASP1 gene encodes the MASP3 protein through alternative splicing. This strain was a knock-in model generated by homologous recombination with an hMASP3 cassette at Masp1 gene locus.

Validation Data

Fig.1 Detection of MASP3 expression in adrenal by RT-PCR. Mouse Masp1 mRNA (154 bp) was detectable only in WT C57BL/6 mice. Human MASP1 mRNA (186 bp) was detectable only in HO hMASP3 knockin mice but not in WT mice. Adrenal RNA was extracted from 6-week-old female wild-type C57BL/6 (WT)(n=2) and homozygous hMASP3 knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of MASP3 expression in hMASP3 mice by WB. MASP3 was detectable in liver from both WT C57BL/6 and HO hMASP3 mice , as the antibody was cross-reactived between human and mouse MASP3. The liver tissue lysates were collected from 7-week-old female wild-type C57BL/6 mice and 7-week-old female homozygous hMASP3 mice, and then analyzed by western blot with anti-MASP3 antibody.

Abbr. HO, homozygous; WT, wild type.

Fig.3 Detection of mouse CFD expression in serum of WT C57BL/6 mice and HO hMASP3 mice by ELISA. CFD was detectable in serum from both WT C57BL/6 and HO hMASP3 mice. Serum were collected from wild-type C57BL/6 mice (n=2, female, 7 weeks old) and HO hMASP3 mice (n=2, female, 7 weeks old), and analyzed by ELISA with anti-mCFD ELISA kit.

Abbr.  HO, homozygous; WT, wild type.
