Fig.1 Detection of mouse CFH expression in serum of WT C57BL/6 mice, HE Cfh-KO mice and HO Cfh-KO mice by ELISA. Compared with WT C57BL/6 mice, CFH levels were significantly decreased in heterozygous Cfh-KO mice and were barely detectable in homozygous Cfh-KO mice. Serum were collected from wild-type C57BL/6 mice (n=3, 6 weeks old), HE Cfh-KO mice (n=6, 6 weeks old) and HO Cfh-KO mice (n=3, 6 weeks old), and analyzed by ELISA with anti-mCFH ELISA kit.
Abbr. HO, homozygous; WT, wild type; HE, heterozygote.

Fig.2 Detection of CFH expression in CFH-KO mice by WB. Mouse CFH was present in the eyes of WT C57BL/6 mice but entirely absent in homozygous Cfh-KO mice. The eye tissue lysates were collected from 6-week-old wild-type C57BL/6 mice, 6-week-old heterozygote CFH-KO mice and 6-week-old homozygous CFH-KO mice, and then analyzed by western blot with anti-CFH antibody.
Abbr. HO, homozygous; WT, wild type.

Fig.3 Detection of mouse C3 expression in serum of WT C57BL/6 mice, HE Cfh-KO mice and HO Cfh-KO mice by ELISA. Compared with WT C57BL/6 mice, C3 levels were significantly decreased in heterozygous Cfh-KO mice and were undetectable in homozygous Cfh-KO mice. Serum were collected from wild-type C57BL/6 mice (n=3, 6 weeks old), HE Cfh-KO mice (n=6, 6 weeks old) and HO Cfh-KO mice (n=3, 6 weeks old), and analyzed by ELISA with anti-mC3 ELISA kit.
Abbr. HO, homozygous; WT, wild type; HE, heterozygote; N.D. not detected.

Fig 4. Frequency of leukocyte subpopulations in the blood. Cfh-KO mice showed a normal immune phenotype in blood. Blood was collected from WT C57BL/6 mice and Cfh-KO (C57BL/6) mice (6-week-old, male, n=3). Flow cytometry analysis of frequencies of leukocyte subpopulations in the blood. Bar indicated expressed as mean ± SEM.
Abbr. HO, homozygous; WT, wild type.

Fig 5. Frequency of leukocyte subpopulations in the spleen. Cfh-KO mice showed a normal immune phenotype in spleen. Spleen was collected from WT C57BL/6 mice and Cfh-KO mice (6-week-old, male, n=3). Flow cytometry analysis of frequencies of leukocyte subpopulations in the spleen. Bar indicated expressed as mean ± SEM.
Abbr. HO, homozygous; WT, wild type.
