Model Description
Validation Data

Fig.1 Detection of GLP1R expression in lung by RT-PCR. Rat Glp1r mRNA (214 bp) was detectable only in wild-type SD rat. Human GLP1R mRNA (329 bp) was detectable only in homozygous hGLP1R(SD,2) knockin rat but not in wild-type rat. Lung RNA was extracted from 6-week-old female wild-type SD rats (n=3) and homozygous hGLP1R(SD,2) knock-in rats (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of human GLP-1R expression in hGLP1R(SD,2) rats by IHC. Human GLP-1R was exclusively detectable in pancreatic islets and brain from homozygous hGLP1R (SD,2) knock-in rats but not in WT rats. Tissues were collected from 6-week-old female WT SD rats and HO hGLP1R(SD,2) rats, then analyzed by IHC. Black arrows indicate positive staining (brown). The scales are same in each image. Magnification, 20X.
Abbr. HO, homozygous; WT, wild type
